The test can be run manually by nut.def configuration as you did.
And DNS tester can be run over IPv4 and IPv6.
I guess that you want to test over IPv6, right?
For IPv6 testing, you need following addresses to be configured.
Your printer:
address: 3ffe:501:ffff:100::10
default gateway: 3ffe:501:ffff:100::1
Tester:
address #1: 3ffe:501:ffff:100::1
address #2: 3ffe:501:ffff:100::20
address #3: 3ffe:501:ffff:100::30
address #4: 3ffe:501:ffff:100::31
address #5: 3ffe:501:ffff:100::32
address #6: 3ffe:501:ffff:101::10
address #7: 3ffe:501:ffff:101::11
address #8: 3ffe:501:ffff:101::20
address #9: 3ffe:501:ffff:101::30
address #10: 3ffe:501:ffff:101::40
address #11: 3ffe:501:ffff:101::50
